Analysis of Prostaglandin E 2 Effect on T Lymphocyte Activation

چکیده

We have investigated the inhibitory potential of prostaglandin E2 (PGE2) with respect to intracellular messengers implicated in the signaling system ofT-lymphocyte activation pathway. Using the fluorescent indicator Quin 2, it is demonstrated that PGE2 inhibits the increase in cytosolic-free calcium concentration ICa211. Reconstitution of calcium mobilization in the presence of PGE2 by the calcium ionophore A23187 results in a partial restoration of both interleukin 2 (IL2) production and cell proliferation and has no effect on the inhibition of transferrin receptor expression. In contrast, the treatment of cell cultures with the tumor promotor 12.0 tetra decanoyl phorbol-13-acetate (TPA) abrogates the suppressor activity of PGE2. When T lymphocyte stimulation is provided by the combination of A23187 and TPA, the PGE2 inhibitory effect does not occur. These data also indicate that the down regulation of transferrin receptor by PGE2 is proximal to protein kinase C activation and is not associated with decreased expression of the functional IL2 receptor.

EFFECT OF TOXOCARA CANIS SECOND STAGE LARVAE ON THE HOST IMMUNE SYSTEM

We have studied functional alteration of immunocytes in mice following inoculation of Toxocara canis second stage larvae. Results indicated depression of lymphocyte blastogenesis in response to concanavalin-A, phytohemagglutinin and allogeneic non B-cells; however, B-cell polyclonal activation was not affected as indicated by the production of total IgG. In contrast, frequency of antibody ...

Adverse Effect of T-2 Toxin and the Protective Role of Selenium and Vitamin E on Peripheral Blood B lymphocytes

Background: T-2 mycotoxin belongs to the Trichothecene family and has damaging effects on the immune system. Objective: To investigate the toxic effect of T-2 toxin on the percentage of peripheral blood B lymphocytes and the potential protective role of selenium and vitamin E. Method: Frequencies of B lymphocytes (CD19+ ) were analyzed after injection of sublethal doses of T-2 toxin into Balb/c...

O 1: The Effects of Vitamin D Supplementation on the T cell Compartment in Multiple Sclerosis

Multiple sclerosis (MS) is a complex neurological disease and its prevalence is about 2 million in the world. Neuroinflammation plays a key role in MS. Vitamins are essential nutrients that have effective role on immune system including activation of lymphocyte and differentiation of T-helper cell. Vitamin D is a micronutrient that is effective on immune function. Deficiently of Vitamin D is a ...

Rat Mesenchymal Stromal Cells Inhibit T Cell Proliferation but Not Cytokine Production Through Inducible Nitric Oxide Synthase

Mesenchymal stromal cells (MSC) have important immunomodulatory properties, they inhibit T lymphocyte allo-activation and have been used to treat graft-versus-host disease. How MSC exert their immunosuppressive functions is not completely understood but species specific mechanisms have been implicated. In this study we have investigated the mechanisms for rat MSC mediated inhibition of T lympho...
